Maintaining Process Consistency And Performance During N-1 Perfusion Scale-Up
Maintaining consistent culture conditions becomes increasingly difficult as intensified upstream processes move from bench to pilot scale. An N-1 perfusion process for a CHO-based monoclonal antibody was evaluated at 5 L, 50 L, and 500 L across GSK and Thermo Fisher development sites, covering seed expansion through intensified fed-batch production. Cultures remained stable at each scale, with reproducible cell expansion and closely aligned growth and mAb production profiles. Viable cell densities approached 1 × 10^7 cells/mL, while gas-liquid mass transfer supported performance at higher densities.
Explore the data to see how performance compared across all three scales.
