A customer sought to replace a chemically defined CHO medium with a more efficient medium and feed pairing strategy in bioreactors. A resulting study, conducted by Thermo Fisher Scientific, showed that the amount and timing of peptone feed impact the product titer. A basal medium and feeds were developed and tested with various peptones to enhance performance. The results showed improvements when basal mediums were used in Gibco™PD-Express Services. An optimized feed strategy resulted in a 70% increase in titer over the control in this study. Adding Gibco™ Difco™ TC Yeastolate ultra-filtered to the feed led to a more than 100% improvement over feeds designed based on spent media analyses. The process was successfully scaled to a bench-scale bioreactor.
