This application note discusses the evaluation of the Corning CellCube System, a platform for adherent cell culture, using a 3L Corning Disposable Spinner Flask in a cell culture incubator. The study describes the setup and process for cell expansion in the CellCube system using the spinner flask as a substitute for a bioreactor controller. The system utilizes a peristaltic pump for circulation and is placed in a cell culture incubator for passive oxygenation, pH buffering, and medium aeration. The study found that the CellCube system was comparable to the control U-shaped flask in terms of cell confluence and demonstrated a "plug-and-play" setup for the system. The 3L Disposable Spinner Flask is capable of supporting medium mixing within a volume range of 300 to 3000 mL and can accommodate medium volume-to-surface area ratios suitable for many cell culture applications. The study also compared cell growth characteristics in the CellCube incubator setup to a traditional bioreactor-controlled CellCube system and found them to be comparable. Understand how the Corning CellCube system, with the use of a 3L disposable spinner flask, can support cell expansion in a standard cell culture incubator.
