Automated Manufacturing Of CD34-Enriched Cell Products From Cryopreserved Mobilized Apheresis On The CliniMACS Prodigy®

Poor graft function after allogeneic stem cell transplantation and prolonged cytopenia following CAR T-cell therapy can leave clinicians searching for effective ways to restore hematopoiesis. One approach is the use of CD34+ stem cell boosts, but fresh donor material is not always available when treatment is needed. This application note explores an automated workflow for generating CD34-enriched cell products from cryopreserved mobilized apheresis material using the CliniMACS Prodigy. The process integrates DMSO removal directly into cell enrichment, eliminating the need for a separate wash step and reducing hands-on manufacturing time. Across multiple runs, the workflow produced highly pure, viable CD34+ cell products with strong cell recovery and substantial T-cell depletion.
