The CliniMACS Prodigy®: An Adaptable, Intelligent Centerpiece For Customizable Manufacturing Strategies

A persistent assumption in cell therapy manufacturing holds that automated processing platforms and third-party expansion technologies operate as separate, incompatible systems. This application note directly challenges that view. Using a CD19/CD20 CAR T cell manufacturing protocol, data from an integrated closed-system workflow demonstrates that combining automated liquid handling with external bioreactor expansion produces cell yields comparable to internal controls, with naïve and central memory phenotypes exceeding 85% for both CD4+ and CD8+ populations, and greater than 90% target cell killing at a 10:1 effector-to-target ratio. Beyond the case study findings, the piece examines how open-architecture design and custom workflow programming enable manufacturers to scale vertically for allogeneic batch volumes, scale horizontally through parallel processing to improve instrument ROI, and maintain GMP-compliant sterility throughout.
