S-SELeCT™ One-Step Large Knock-In Technology
Simple, scalable genome engineering at the naturally occurring S-SELeCT™ safe harbor locus.
A new generation of genome engineering tool
Designed to enable gene insertions when you can’t engineer in a landing pad, S-SELeCT™ technology—short for Site-Specific, Efficient, Large Cargo Targeting—is the newest genome editing innovation from the team that developed the TARGATT™ large knock-in platform.
S-SELeCT at a glance
At Applied StemCell, we believe that ensuring the safety of the next generation of cell and gene-based medicines means developing precision gene insertion tools that integrate site specifically and without introducing free DNA breaks.
This is why we developed the S-SELeCT™ platform.
Consisting of an evolved serine integrase, an optional catalytically inactive Mad7 fusion (dMad7), a donor plasmid with your gene-of-interest, and a proprietary safe harbor locus that naturally exists in the human genome, S-SELeCT™ technology is a simple, safe, scalable genome engineering platform.
- Simple one-step gene insertion—no need to pre-insert a landing pad
- Lean, two- to three-component system—evolved S-SELeCT™ integrase, optional dMad7 fusion, donor plasmid with your gene-of-interest
- Large cargo compatible
- Site-specific
- Patented, naturally existing human safe harbor locus, the S-SELeCT™ site
- Negligible off-target events
Directed evolution for efficiency and specificity

Our evolved S-SELeCT™ integrase is specific for a naturally existing safe harbor locus in the human genome, which we identified through analysis of gene expression data and genome-wide scans.
