To address process-specific challenges in PSC manufacturing, the team scaled iPSC expansion from a 0.1-liter to an 80-liter bioreactor. A key challenge was optimizing the seed train culture, which is critical for consistent 3D expansion. A simplified 2D seed train with two passages was developed, focusing on maintaining key process input variables (PIVs) like inoculation density and culture duration.
