Human Platelet Lysate (hPL) Improves Expansion Of MSCs

Mesenchymal stromal/stem cells (MSCs) are critical to cell-based therapeutic development due to their immunomodulatory properties. Traditionally cultured in serum-supplemented media, human platelet lysate (hPL) offers a superior alternative, promoting cell proliferation and migration thanks to higher concentrations of growth factors like PDGF, TGF-β, and IGF. Additionally, hPL can support cell adhesion, potentially removing the need for surface-coating proteins like fibronectin.
A study investigated the effect of supplementing a xeno-free, serum-free medium with varying concentrations of hPL (0.5% to 5%) on MSC expansion. The results showed that adding hPL significantly increased Population Doubling Levels (PDLs) in a dose-dependent manner compared to the no-hPL, fibronectin-coated control. Furthermore, any concentration of hPL tested eliminated the need for fibronectin precoating to achieve MSC adhesion and improved proliferation. Cell viability and surface marker expression remained consistent across all hPL-supplemented conditions, demonstrating that hPL supports robust expansion without altering cell phenotype.
