How-To Guide For TARGATT™ Transgenic Kit, By Dr Ruby Chen-Tsai

The TARGATT™ Transgenic Kit enables rapid and precise generation of site-specific knock-in transgenic mice at defined chromosomal loci, offering a more efficient alternative to traditional methods. Conventional techniques such as pronuclear microinjection or lentiviral injection often result in random transgene insertion, which can cause position effects that lead to transgene silencing or disruption of endogenous gene expression.

Additionally, random insertion may introduce multiple copies of the transgene, resulting in genomic instability at the insertion site. The TARGATT™ system overcomes these challenges by using a proprietary site-specific DNA integration technology combined with genetically engineered TARGATT™ mice (strain code 549). This approach ensures the transgene is inserted precisely at the desired locus, guaranteeing stable and consistent gene expression. The streamlined process significantly reduces the time required to generate transgenic mouse models, enabling researchers to obtain their desired knock-in models in as little as three months.

This innovation greatly accelerates the timeline for functional studies, disease modeling, and therapeutic research by providing genetically uniform and reliable transgenic animals, free from the drawbacks associated with random insertion techniques.
