This study investigates the efficiency and effectiveness of various lysis methods for Adeno-Associated Virus (AAV) recovery, focusing on optimizing capsid and vector genome (Vg) yields across multiple AAV serotypes. Key approaches evaluated include mechanical lysis, freeze-thaw cycles, single- and multiple-pass sonication, and the application of both commercial and chemical lysis reagents. Recovery rates were normalized to provide clear comparisons of capsid integrity and genome preservation under different lysis conditions and reagent concentrations.

The findings reveal significant variability in performance, with specific reagents and methods showing superior recovery rates for distinct AAV serotypes. These insights are critical for researchers aiming to maximize yield and quality in AAV production workflows, particularly in therapeutic applications where efficiency and reliability are paramount.
