AAV-based gene therapy can be compromised by the presence of empty capsids, which can also obstruct vector purification. Mass photometry is an effective tool for characterizing these AAVs, as it can quickly and accurately measure the empty/full AAV ratio for capsids of any serotype using very little sample. It can also quantify partially filled and overfilled AAV capsids and provide a rough titer estimate. In a webinar, Santoshkumar Khatwani from Sangamo Therapeutics discussed his work on developing an analytical workflow for gene therapy products. James Wilkinson from Refeyn then explained the functioning of mass photometry and introduced new advancements in the SamuxMP product line. These include an automation upgrade and upcoming GMP-compliant software, making mass photometry an ideal solution for manufacturing and QC environments.
