We can help you with molecular cloning services, from gene synthesis, mutagenesis, and shRNA cloning into any vector backbones, including AAV, lentiviral, adenoviral, and retroviral vectors. All you have to do is provide us with the gene sequence, the name of your preferred promoter, and the vector backbone, and in turn, we will provide you ready-to-use plasmids or packaged viral vectors.

Molecular Cloning Services

As experts in AAV, adenovirus, and lentivirus molecular cloning services, we can tackle your molecular cloning project to save you time and effort in the lab. For your convenience, we offer everything from designing and constructing custom viral vector to packaging and purifying viral particles. Simply provide us your project details and we will deliver ready-to-use viral particles generated according to your exact specifications.

Gene Synthesis and Mutagenesis

We offer gene synthesis services and site-directed mutagenesis services as a time-saving way to generate the construct that meets your exact specifications, creating specific variants or disease specific mutants. Simply send us your desired sequence for gene synthesis, or choose from our extensive collection of human ORFs as templates for site-directed mutagenesis. Our team of scientists are able to do everything from synthesizing or mutating a gene, to cloning into a viral vector backbone or any other vectors, to packaging and purifying viral particles that are ready for in vitro or in vivo gene delivery.

shRNA Cloning

We offer multiple shRNA cloning options to help you plan the most time-saving and cost-effective strategy for your gene silencing experiments. For most customers, we recommend performing initial testing with shRNAs in a viral vector backbone (i.e., transfecting with plasmid and validate in vitro). We guarantee at least 70% knockdown of your gene of interest with our custom designed shRNA constructs. Once you have determined the best shRNA for your experiments, we can package the construct to generate AAV shRNA, adenovirus shRNA, or lentivirus shRNA viral particles for efficient gene knockdown.

Frequently Asked Questions (FAQs) for Molecular Cloning Services

Are there any limitations to which ORFs can be shuttled?

If you want to generate viral particles, you must not exceed the packaging capacity of the virus. Please note that the packaging capacity refers to the total insert size between and including the ITRs or LTRs.

  • AAV cloning capacity: 4.7 kb
  • Adenovirus cloning capacity: 7.5 kb
  • Lentivirus cloning capacity: 6 kb

What information do I need to provide for gene synthesis services?

For gene synthesis services, please provide the following:

  • Gene ID/Name (specify variant, if applicable) or file containing the desired sequence
  • Name of desired viral vector backbone or name of customer-provided vector

What information do I need to provide for mutagenesis services?

For mutagenesis services, please provide the following:

  • Gene ID/Name (specify variant, if applicable) or file containing the desired sequence
  • Description of sites/base pairs to mutate
  • Name of desired viral vector backbone or name of customer-provided vector
  • If you are sending your own vector for mutating or cloning, please send us at least 5 ug and a sequence/plasmid map

Does Charles River validate shRNA sequences?

We do not validate shRNA sequences designed as a part of the shRNA molecular cloning services. However, we will send you the shRNA plasmids to test. Once you determine which shRNA construct works best for your experiments, we can package your selected shRNA construct into virus.
