Behind The Data: Engineering Hepatocyte-Targeted LNPs For RNA And DNA Delivery To Primary Human T Cells
As gene therapy advances toward larger and more complex payloads, delivery methods must evolve with it. ElevateBio’s lipid nanoparticle (LNP) platform is designed to deliver both RNA and DNA to primary human T cells, offering an alternative to viral vectors and electroporation. In comparative testing, LNPs achieved high mRNA transfection while preserving cell viability, sustained CD19-CAR expression, and enabled TRAC knockout at levels comparable to electroporation. An optimized DNA LNP also produced more than 90% GFP-positive cells at a relatively low dose. When RNA and DNA LNPs were combined for large serine recombinase-mediated CAR insertion, efficiency exceeded 85%, compared with about 20% using electroporation.
