Advancing mRNA Therapeutics: Lipid Nanoparticles Demonstrate Favorable Safety And Efficacy Profile With Repeated mRNA Administration In Normal And Tumor Bearing Mice

Can mRNA-LNP formulations remain safe and effective through repeated administration? Researchers evaluated multiple formulations in mice across as many as nine doses, administered at 21-day intervals over approximately six months. The analysis tracked clinical observations, liver enzymes, inflammatory markers, anti-PEG antibodies, and antigen-specific immune responses to build a detailed picture of repeat-dose performance. Novel LNP formulations were associated with minimal adverse events and no indications of systemic toxicity. In a melanoma mouse model, a lead formulation also demonstrated antitumor effects following four repeated doses.
