A Robust System For High-Titer, Platform-Agnostic AAV Production In HEK293 Cell Lines

Improving viral titer and full capsid ratios in AAV production means optimizing many variables at once, yet the medium underlies all of them.
Scientists from Lonza Walkersville, including Leah Hirsh, Katharyne Bostin, and Samin Rezania, evaluate a chemically defined, animal-origin-free medium system against three criteria: compatibility with fixed components like serotype and plasmids, flexibility with variable ones like cell line, transfection reagent, and enhancer, and strong baseline performance. Data span AAV2, AAV8, and AAV9, multiple HEK293 cell lines, and a move from shake flasks to 3 L bioreactors with downstream purification. A supplement-driven titer increase of roughly 3x is reported, along with cell growth and viability data.
