A Robust Immune Monitoring Spectral Cytometry Assay With Six Open Channels For Biomarker Flexibility

For biomarker development, cytometry assays offer advantages over other multiplex methodologies due to real-time sample analysis, lower cost, and comparatively rapid data turnaround. However, the complexity of spectral cytometry panel development limits the adaptation of established assays. Cytometry assays can also be undermined by poor reproducibility for rare populations, reduced resolution after fluorescence spill-over correction, and non-optimized data analysis practices.

A pan-immune monitoring spectral cytometry assay was developed on the Cytek Aurora for cross-program applications, while opting for six additional drop-in channels such that biomarkers of interest could be incorporated. This study emphasizes why optimized reference control practices are an absolute requirement for successful application of spectral cytometry assays.
