A New 30-Color Spectral Flow Cytometry Panel For T Cell Therapy Phenotypic Characterization
By David Draper, Brogan Yarzabek, Stacey Roys, Anita J. Zaitouna, Philip Lapinski, Amber Rowse and Scott Wise; Labcorp, Ann Arbor, MI

Spectral flow cytometers offer advanced immunophenotypic analysis with panels that can exceed 30 colors, providing deep insights into the immune response. Labcorp has developed a 30-color immunophenotyping panel using the Cytek Aurora™ Spectral Flow Cytometer, designed to deliver comprehensive immune profiling. The panel includes a backbone of one viability dye and ten lineage markers that allow for the profiling of T cell, NK cell, and B cell subsets. Additionally, 19 functional markers are included, enabling the characterization of activation, exhaustion, cytotoxicity, proliferation, and memory T cell subsets.
The data presented demonstrates how spectral flow cytometry can be applied to assess T cell therapies in various studies, ranging from in vitro assessments and preclinical tumor-bearing mouse models to clinical trials.
