Opto™ Cell Therapy Development Workflow: Single Cell Cytokine Assay
Adoptive T cell therapies have shown greatest clinical efficacy when the transferred T cells exhibit cytokine polyfunctionality, efficient target cell killing, and proliferation in response to antigens, leading to persistence of the transferred cell. However, many methods for defining T cell phenotypes fail to measure multiple parameters on individual T cells, forcing scientists to draw conclusions about samples that can be skewed by the extreme heterogeneity usually observed in bulk T cells measurements.
With the Opto Cell Therapy Development workflow, user-specific cell-cell interactions can be rapidly and precisely assembled. Hundreds to thousands of experiments with single T cells can be run in parallel. By adding multiplexed functional assays, a rich and robust visual record of T cell phenotype and function can be generated. This enables use cases such as:
- CAR-T construct screening and validation
- Antigen or TCR discovery and validation
- Investigating helper or regulatory T cell function
